Method: Create a multiple position timelapse experiment moving across diagonals to record accuracy of stage movement in both x and y, moving to defined landmarks at each point. Record the deviation from original point with each subsequent return.
Justification: Any movement quantification of a sample requires knowing that there is no (or a pre-known) amount of accuracy in the stage movement. This improves data quality and aids reproducibility.
Requirements: A marked slide or dish securely positioned within the motorised stage plus an analysis method to record x and y deviation between timepoints, e.g. ImageJ or Argolight Daybook.
